The two car crash happened on the Mountain Road between Buncrana and Carndonagh at around 10.50 last night.
A man in his 40's who was driving one of the cars was pronounced dead at the scene.
The driver and passenger of the second vehicle were taken to Letterkenny University Hospital, where the condition of the female passenger is described as 'serious'.
The Mountain Road is currently closed between Buncrana and Carndonagh for a forensic examination, with local diversions now in place.
Gardaí are appealing for witnesses to come forward, especially anyone who may have dash-cam footage.
